Cue Biopharma (NASDAQ: CUE)'s CUE-501, a molecule that binds to a B-cell-specific membrane protein while selectively engaging virus-specific memory killer T cells to deplete targeted B cell, has triggered a USD 7.5 million preclinical milestone payment under a collaboration and license agreement with Boehringer Ingelheim. The payment follows Boehringer Ingelheim's selection and approval of its first compound for lead optimization under the multi-year agreement. Cue expects to receive the payment in May 2026.

The milestone is a follow-on event under the original collaboration signed in April 2025, which carried a USD 12 million upfront payment and up to USD 345 million in additional research, development, and commercial milestone payments, plus royalties on net sales.

Deal context

CUE-501 is built on Cue's proprietary Immuno-STAT platform, which the company describes as generating injectable bispecific fusion proteins and T cell engagers engineered to simultaneously bind a cell-surface marker on a specific immune cell population and deliver a targeted immunomodulatory signal to that cell. In the context of CUE-501, the intended effect — as described by Cue — is selective engagement of memory T cell subsets to drive depletion of pathogenic B cells implicated in autoimmune and inflammatory diseases. The bispecific architecture is designed to enforce selectivity through co-engagement geometry, limiting activity to cells that co-express both targeted features.

The approach is mechanistically distinct from pan-B cell depletion strategies such as anti-CD20 antibodies, which eliminate the broader B cell compartment. CUE-501 remains at the preclinical lead optimization stage; no clinical trial has been initiated for this asset.
