Switzerland-based Lonza (SIX: LONN) has granted the UK's Engitix Therapeutics an exclusive license to develop one undisclosed target using its antibody-drug conjugate (ADC) technology suite, marking the latest in a rapid succession of ADC development deals signed by the CDMO since the start of 2026. Under the agreement, Engitix gains access to Lonza's GlycoConnect, HydraSpace and SYNtecan E linker-payload technologies to build an antibody-drug conjugate directed at a target identified through its proprietary extracellular matrix (ECM) discovery platform.
Engitix, which has built its pipeline around ECM-based drug discovery rather than conventional cell-surface target identification, will assume full responsibility for research, development, manufacturing and commercialization of the resulting ADC. Lonza will be responsible for manufacturing the proprietary conjugation and linker-payload components, and is eligible for upfront, clinical, regulatory and commercial milestone payments plus royalties on net sales. Neither the specific financial terms nor the target antigen were disclosed.
The licensed package combines three components that address distinct engineering problems in ADC construction. GlycoConnect exploits the conserved glycan at the antibody's Asn297 site to attach payloads with a defined drug-to-antibody ratio, enabling site-specific conjugation with a defined drug-to-antibody ratio. HydraSpace, a polar spacer, counters the aggregation and rapid clearance problems associated with hydrophobic cytotoxic payloads. Engitix's contribution sits upstream. The company's human ECM platform generates target hypotheses from disease-derived tissue rather than animal models or conventional surfaceome screening, with a stated focus on fibrosis, oncology and other conditions driven by pathological tissue remodeling. The Lonza deal marks Engitix's application of that target-discovery engine to the ADC modality specifically — pairing tumor-selective targets identified in the extracellular matrix with a validated bioconjugation toolkit rather than developing conjugation chemistry internally.
The partnership is the latest in a string of deals signed by Lonza in relation to ADC development, including a multi-target agreement with Cambridge, Massachusetts-based Stipple Bio on June 4 and an exclusive, target-specific license to San Diego-based Antharis Therapeutics for dual-payload ADCs in gastrointestinal cancers on June 15. Both deals used the identical upfront-milestone-royalty structure now applied to Engitix.
