Insilico Medicine revealed that the novel small molecule MEN2501, developed through Insilico’s generative AI drug discovery platform, has been dosed in a first patient in a Phase I first-in-human clinical trial. The news triggers a USD 5 million milestone payment from Menarini Group to Insilico, under the terms of their licensing deal focused on the drug candidate signed in January 2025.

MEN2501 (previously ISM9682) is a highly differentiated small molecule inhibitor of kinesin KIF18A, a kinesin motor protein critical to chromosome alignment during cell division. By disrupting this mechanism, the molecule aims to selectively induce cell death in cancer cells with high chromosomal instability. The Phase I trial is being managed by Menarini Group subsidiary Stemline Therapeutics, and is targeting patients with chromosomally unstable solid tumors, particualrly ovarian cancer.

The first-in-human trial positions MEN2501 within an emerging field of mitotic mechanism-targeting therapies. With total potential agreement value exceeding USD 550 million, the molecule represents a significant bet on AI-discovered oncology treatments.
