TAG1 and Dana-Farber collaborate on Lead-212 supply for targeted alpha therapy research

St. Louis-based TAG1 Inc., and the Molecular Cancer Imaging Facility (MCIF) at Dana-Farber Cancer Institute in Boston announced a strategic collaboration under which TAG1 will supply its proprietary Lead-212 (Pb-212) generator to Dana-Farber’s MCIF for use in preclinical and translational research on targeted alpha therapies.

The deal is structured as a research supply and collaboration arrangement rather than a traditional licensing transaction. No specific drug candidates are named in the agreement; instead, the collaboration centers on TAG1’s portable generator platform as an enabling technology for radiopharmaceutical development across multiple potential oncology targets. Financial terms were not disclosed.

TAG1 will provide its Pb-212 generator to Dana-Farber’s MCIF, giving researchers on-site access to Lead-212 for conjugation to targeting molecules such as antibodies, peptides, or small molecules. The collaboration is designed to support both early-stage discovery and translational research and is expected to underpin a portfolio of research programs. The arrangement also creates a framework for external academic and industry partners developing targeted alpha therapies to access Dana-Farber’s scientific infrastructure alongside TAG1’s isotope supply capabilities.

Deal context: lead-212 uses

Lead-212 is an alpha-emitting radioisotope with a half-life of approximately 10.6 hours. In alpha particle therapy cancer applications, Pb-212 is conjugated to tumor-targeting ligands that deliver high-energy alpha particles directly to cancer cells, inducing double-stranded DNA breaks with a short tissue path length that limits damage to surrounding healthy tissue. This mechanism differentiates alpha-emitting radiopharmaceuticals from beta-emitting agents such as Lutetium-177, which have a longer range and lower linear energy transfer. Lead-212 also forms a theranostic pair with Lead-203, a gamma emitter suitable for SPECT imaging, enabling matched diagnostic and therapeutic development.

The most advanced Pb-212 clinical program is 212Pb-DOTAMTATE (AlphaMedix), developed by Perspective Therapeutics in collaboration with MD Anderson Cancer Center, which targets somatostatin receptor type 2 (SSTR2) in neuroendocrine tumors. That program is in Phase I/II testing under NCT05636618. A separate Phase I study of 212Pb-Pentixather, targeting CXCR4 in pulmonary neuroendocrine tumors, is registered under NCT05557708. TAG1’s generator does not itself constitute a drug candidate but serves as the supply infrastructure required to produce Pb-212 on-site, addressing a logistical constraint imposed by the isotope’s short half-life.

For TAG1, the Dana-Farber collaboration is the latest in a series of partnerships announced in 2025 and 2026 as the company scales its commercial relationships. TAG1 previously entered a strategic partnership with PharmaLogic to advance decentralized Lead-212 generator development, and expanded a partnership with NRG PALLAS to enable European access to its Pb-212 generator and broaden Radium-224 supply. The TAG1 Dana-Farber collaboration extends the company’s institutional footprint to a second major US academic cancer center, complementing the existing MD Anderson axis for Pb-212 clinical research. No prior relationship between TAG1 and Dana-Farber was identified before this announcement.

Dana-Farber’s MCIF functions as a preclinical radiopharmaceutical R&D core supporting both internal and external partners in radiochemistry optimization and in vivo model generation. The facility’s engagement with TAG1 represents a new capability addition for molecular cancer imaging and next generation radioisotopes research at the institute. Anthony Belanger, Scientific Director of the MCIF, stated that the facility’s teams will work with TAG1 to support academic scientists and industry partners at Dana-Farber.

The Pb-212 supply landscape is concentrated among a small number of producers. Orano Med, a French company, controls much of the global thorium-228/lead-212 generator capacity and has supplied Perspective Therapeutics for its clinical programs. Orano Med has also collaborated with RadioMedic at the Czech Academy of Sciences to expand Pb-212 production capacity. TAG1’s proprietary generator represents an alternative supply source, and its portable design is intended to enable decentralized, on-site isotope production. Broader industry activity around targeted alpha therapies and radiopharmaceutical oncology has increased. Bayer has maintained interest in alpha-emitting platforms since its commercialization of Xofigo (radium-223) and has explored Pb-212 conjugates alongside its actinium-225 work. Lantheus Holdings has been reported as exploring partnerships around Pb-212-based assets as it expands beyond diagnostic radiopharmaceuticals into therapeutic oncology. Novartis, through its Advanced Accelerator Applications subsidiary, commercializes Lutathera (177Lu-DOTATATE), the approved beta-emitting SSTR2-targeted radioligand therapy for gastroenteropancreatic neuroendocrine tumors, against which Pb-212-based alpha therapies are being positioned as a next-generation alternative.
