Congruence raises USD 39.5m to advance protein misfolding correctors to clinic

Congruence Therapeutics, a Montreal-based biotechnology company developing small molecule correctors for diseases of protein misfolding, has closed a USD 39.5 million financing round. The financing will support clinical and preclinical programs spanning genetic obesity, Parkinson’s disease, and alpha-1 antitrypsin deficiency.

The round was co-led by Dimension, a new investor, and OrbiMed, an existing backer. Additional participants included Amplitude Ventures, FSTQ, Lumira, Investissement Québec, BDC Capital’s Thrive Venture Fund, Driehaus, and Silver Arc.

Congruence is currently running a Phase I/Ib clinical trial for CGX-926 in healthy subjects and patients with MC4R-deficient genetic obesity. The firm is also planning to complete IND-enabling work for two additional candidates targeting GBA1 Parkinson’s disease and alpha-1 antitrypsin deficiency, with CTA/IND filings targeted for early 2027.

The lead asset, CGX-926, is a small molecule corrector designed to rescue misfolded melanocortin-4 receptor protein in patients with MC4R genetic obesity, a monogenic condition in which missense mutations cause the receptor to misfold and degrade before reaching the cell surface. The GBA1 and alpha-1 antitrypsin programs apply the same protein misfolding correctors approach to distinct targets with different disease biology.

Congruence’s pipeline is built on its proprietary Revenir platform, a computational drug discovery engine that models the conformational dynamics of proteins across functional states. The platform uses molecular dynamics and machine learning to identify cryptic and allosteric binding sites where small molecule correctors can stabilize a protein’s native fold, enabling it to pass cellular quality control and function normally. The pharmacological chaperoning concept underlying this approach has been validated in cystic fibrosis through Vertex Pharmaceuticals’ CFTR modulators, and Congruence is applying analogous logic to a broader set of genetically validated targets.

Beyond its internal pipeline, Congruence maintains R&D collaborations with large pharmaceutical partners. The company has a multi-target research partnership with Ono Pharmaceutical, announced in 2024 and since expanded, under which Congruence leads discovery of small molecule correctors for targets across oncology, neurology, and immunology. A separate collaboration with an undisclosed global pharmaceutical company focuses on a metabolic target, the company said.

Co-founder and CEO Dr. Clarissa Desjardins leads the company. Congruence previously raised USD 32 million in 2025 to advance CGX-926 into the clinic. The current round brings cumulative disclosed financing to at least USD 71.5 million.
