Soufflé rises with USD 135m Series B to advance FSHD and heart failure therapies

Boston, Massachusetts-based Soufflé Therapeutics, Inc. raised an oversubscribed USD 135 million Series B to fund its first clinical studies for three RNA therapy programs in facioscapulohumeral muscular dystrophy (FSHD), heart failure, and muscle and metabolic disorders.

The round was led by Juvia Group and added new institutional investors TCG Crossover Management, BB Biotech, Fidelity Management & Research Company, Balyasny Asset Management, and an unnamed sovereign wealth fund. Existing investors (including Amgen Ventures, ARCH Venture Partners, Leaps by Bayer and others) also participated. Soufflé previously closed a USD 200 million Series A in October 2025, led by Bessemer Venture Partners.

Soufflé's REMEDY (Receptor Mediated Delivery) platform is designed to deliver oligonucleotide therapies to selected cell types outside the liver using proprietary receptor-binding ligands. Its lead candidate, SFL-0821, is an antibody–siRNA conjugate designed to suppress DUX4 expression in skeletal muscle for FSHD, with a Phase I/II study (NCT07798609) registered to evaluate safety, tolerability, and preliminary efficacy. A second program targets the phospholamban (PLN) gene in cardiomyocytes for heart failure, while a third addresses an undisclosed target in muscle and metabolic disorders.

The company has established pharmaceutical partnerships with AbbVie, Amgen, Bayer, and Novo Nordisk. In January 2026, Bayer and Soufflé announced a strategic collaboration and global licensing agreement to develop a cardiomyocyte-targeted siRNA therapy for a form of dilated cardiomyopathy, with financial terms undisclosed.
