Roche taps Atavistik Bio in USD 1.9bn allosteric drug discovery deal

Cambridge, Massachusetts-based Atavistik Bio entered a strategic research collaboration with Roche (OTCQX: RHHBY) to discover small-molecule therapies against multiple undisclosed cardiovascular, renal, and metabolic disease targets. Atavistik will receive USD 70 million upfront and could earn up to USD 1.9 billion in research, development, and commercial milestones, plus tiered royalties.

The deal gives Roche access to Atavistik’s AMPS allosteric drug-discovery platform, which is designed to identify functional binding pockets on disease targets that have been difficult to modulate with conventional small molecules. Atavistik will lead early discovery and hit-to-lead work, with Roche assuming responsibility for preclinical and clinical development, regulatory activities, and commercialization.

The collaboration extends Atavistik’s platform beyond its internally focused hematology and vascular-disease pipeline. The company’s lead asset, ATV-1601, is an oral allosteric AKT1-selective inhibitor in clinical development for hereditary hemorrhagic telangiectasia, while a separate JAK2V617F-selective program is being advanced for myeloproliferative neoplasms.

Atavistik raised a total of USD 160 million in its Series B financing across an initial December 2025 round and a March 2026 extension, with proceeds supporting ATV-1601 and the JAK2V617F program through clinical proof-of-concept.

For Roche, the partnership adds another discovery component to an expanding CVRM portfolio. The company completed its acquisition of 89bio in October 2025 in a deal worth up to approximately USD 3.5 billion, gaining Phase III FGF21 analog pegozafermin for metabolic dysfunction-associated steatohepatitis.
