Vertex Pharmaceuticals (Nasdaq: VRTX) completed its acquisition of Crinetics Pharmaceuticals on September 1, 2026, adding a commercially launched rare endocrine therapy and a Phase III pipeline candidate to its portfolio in a deal that establishes endocrinology as the company's fifth therapeutic pillar alongside cystic fibrosis, hematology, acute pain, and renal disease.

The all-cash transaction, announced in July 2026, valued Crinetics at USD 10.0 billion in total equity, or approximately USD 8.8 billion net of estimated cash acquired, at USD 85.00 per share. Vertex said the deal received the required regulatory clearances and Crinetics shareholder approval before closing. The transaction is expected to be accretive to non-GAAP operating income in 2029, with full accounting and financial details to be provided at Vertex's Q3 earnings call on November 2, 2026.

The commercial anchor is paltusotine (Palsonify), the first once-daily oral somatostatin receptor type 2 (SST2) nonpeptide agonist approved for adults with acromegaly by the US FDA in September 2025, and the European Commission in 2026. By selectively binding SST2 receptors to suppress excess growth hormone and insulin-like growth factor-1 secretion, paltusotine offers an oral alternative to the intramuscular or deep subcutaneous injectable somatostatin analogues that have historically dominated acromegaly management. Another pipeline asset is atumelnant, a once-daily oral adrenocorticotropic hormone (ACTH) receptor antagonist that acts at the melanocortin type 2 receptor (MC2R) in the adrenal cortex, currently in Phase III development for congenital adrenal hyperplasia (CAH) and Phase II development for Cushing's syndrome.
